One of the first steps in examining the safety and efficacy of dietary supplements designed to increase mental acuity, performance, and energy is to characterize the physiological responses to acute administration. Objective: The purpose of this placebo-controlled, double-blind clinical trial was to determine the effects of acute (single-dose) Energize™ supplementation in healthy men and women. Methods : After giving informed consent and being cleared for participation by passing a screening physical, ten healthy men [mean ± SD age, height, weight (36.8 ± 1.2 y, 179.2 ± 2.8 cm, 88.3 ± 16.6 kg)] and ten healthy women (35.2 ± 6.9 y, 169.6 ± 4.3 cm, 61.6 ± 3.9 kg)] completed two trials using a counterbalanced design. Measurements of resting metabolic rate (RMR) and substrate oxidation, assessed via indirect calorimetry (Parvo Medics TrueMax 2400), were made at baseline (pre-ingestion) and for 10 minutes at the end of each hour for two hours post-ingestion; systemic cardiovascular activity (resting heart rate and blood pressure) were measured at 0, 30, 60, 90, and 120 minutes post-ingestion; and Profile of Mood States (POMS) were measured pre and 120 minutes post-ingestion. Results: Two-way ANOVA revealed a significant interaction (p<0.00004) for RMR between conditions. Specifically, ingestion of Energize™ caused a statistically significant increase in RMR at 60 min (11%; from 3.92 ± 0.32 to 4.35 ± 0.22 ml/kg/min, P<0.0001) and 120 min (14%; from 3.92 ± 0.32 to 4.46 ± 0.33 ml/kg/min, P<0.0001) compared to the placebo trial. No changes in heart rate, blood pressure, or POMS were noted between trials. Conclusions: These preliminary findings indicate that a single dose (2 tablets) of Energize™ safely and effectively increases RMR for at least two hours post-ingestion. Future research should: 1) determine how long RMR remains elevated from acute Energize™ administration; and 2) assess the effects of chronic Energize™ supplementation on body weight, body composition, and exercise performance.
